Исследование ассоциации литического нетоза с изменением плазменного уровня ИЛ-8 в эксперименте.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Использовали экспериментальные in vitro модели покоящихся и предварительно активированных к литическому нетозу нейтрофилов, взвешенных в 1 мл аутоплазмы. Все образцы инкубировали четыре часа при 37°С, далее центрифугировали для осаждения клеток, плазму отбирали для измерения содержания ИЛ-8 методом проточной цитометрии.</p></sec><sec><title>Результаты</title><p>Результаты. В плазме, инкубированной с индуцированными к нетозу клетками, концентрация ИЛ-8 в три раза выше, чем в образцах с покоящимися нейтрофилами.</p></sec><sec><title>Заключение</title><p>Заключение. Гибель нейтрофильных гранулоцитов путем литического нетоза ассоциирована с полным высвобождением внутриклеточных запасов ИЛ-8, что сопровождается резким увеличением его концентрации в зоне формирования нейтрофильных внеклеточных ловушек и может способствовать хемотаксису циркулирующих нейтрофильных гранулоцитов и последующей их активации в данном участке с развитием дополнительного протеолитического и свободнорадикального повреждения тканей.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>The aim of the research</title><p>The aim of the research. To verify whether netosis is associated with variation of IL-8 plasma level in vitro.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. We used experimental in vitro models of isolated from whole blood non-stimulated and PMA-activated (100 nm) neutrophils suspended in 1 ml of platelets poor plasma. All samples were incubated for four hours and then were centrifuged at 1500 r.p.m. for 10 min. 50 µl of plasma from each sample were transferred into separate tubes to quantify IL-8 using flow cytometry method.</p></sec><sec><title>Results</title><p>Results. A threefold increase in concentration of IL-8 was revealed in plasma containing PMA-stimulated cells when comparing with non-stimulated samples.</p></sec><sec><title>Conclusion</title><p>Conclusion.
